Phytic acid regulates proliferation of colorectal cancer cells by downregulating NF-kB and β-catenin signalling

Objectives Phytic acid (PYT) also known as inositol hexakisphosphate or polyphosphate has shown a broad range of biological effects including anti-inflammatory, antioxidant, and anticancer in several preclinical studies. This study aimed to investigate the PYT vitro on HCT116 HT-29 cell lines analyse intricate mechanism NF-κB-β-catenin signalling pathways. Material methods Both were treated with PYT, analysed for viability, apoptosis, progression cycle, DNA fragmentation. Gene protein expression analysis was performed assess molecular mechanism. Results suppressed proliferation colorectal cancer dose- time-dependent manner an estimated IC 50 value 2.96 3.35 mm, respectively. caused cycle arrest at G2/M phase both CRC induced mitochondrial intrinsic apoptosis via activation caspase-9 caspase-3 cascade. pro-inflammatory markers especially COX-2 iNOS, IL-lβ, IL-6, IL-10. Analysing behind showed that it levels NF-κB β-catenin inhibited cyclin Dl c-Myc (its downstream targets) COX-2. Conclusion The results collectively indicate potent anti-inflammatory anti-proliferative mediated by downregulating advocate natural supplementation can be effective preventive approach controlling region.
